The Microbiological Determination of Nicotinic Acid, Nicotinamide, and Nicotinuric Acid

چکیده
The nicotinic acid assay most generally satisfactory is the Lactobacillus arabinosus procedure (1, 2) as regards sensitivity and non-interference from other compounds. However, this organism responds to nicotinamide and nicotinuric acid as well as to nicotinic acid. Gaines and Stahly (3) have proposed the use of Leuconostoc mesenteroides for the assay of several members of the vitamin B complex, including nicotinic acid. We have found that this organism responds to nicotinic acid, but not to nicotinamide at low concentrations, nor to nicotinuric acid. By the use of acid hydrolysis of the amide, these two organisms can be used for the determination of all three compounds.

High-performance liquid-chromatographic determination of free nicotinic acid and its metabolite, nicotinuric acid, in plasma and urine.
We report a liquid-chromatographic procedure for determining free nicotinic acid and a metabolite, nicotinuric acid, in plasma and urine. Five-tenths milliliter of urine or deproteinized plasma is evaporated and the residue analyzed isocratically by reversed-phase ion-pair chromatography, with measurement of the eluted nicotinic acid and nicotinuric acid at 254 nm. Nicotinic acid, nicotinuric a...

Several compounds have been described as metabolites of nicotinic acid and nicotinamide in mammals. These are nicotinic acid (I), nicotinamide (2)) nicotinuric acid (2)) trigonelline (2)) W-methylnicotinamide (NMN) (3-6)) and 1-methyl-3-carboxylamide-6-pyridone (7, 8). According to the observations of various workers mammals may be divided into two groups in so far as the metabolism of nicotini...

We investigated a useful chemical index for an excessive nicotinamide intake and how this excessive nicotinamide intake affects the tryptophan-nicotinamide metabolism in rats. Weaning rats were fed on a tryptophan-limited and nicotinic acid-free diet containing no, 0.003%, 0.1%, 0.2%, or 0.3% nicotinamide for 21 days. Urine samples were collected on the last day and analyzed the intermediates a...

The identification of nicotinamide-N1-oxide as a metabolite in the urine of a schizophrenic patient prompted a study of the relative metabolism of nicotinic acid and nicotinamide in mental patients and healthy volunteers. Metabolites quantified included N1-methyl-2-pyridone-5-carboxamide, N1-methyl-4-pyridone-3-carboxamide, N1-methylnicotinamide, nicotinuric acid, and nicotinamide-N1-oxide. Mor...

Nicotinic acid is outstanding among the B vitamins in that its metabolic fate has been very extensively investigated. These studies, however, have been carried out mostly in higher animals, and they have revealed that (i) N'-methylnicotinamide is, in general, the major end-product of the metabolism of both nicotinic acid and nicotinamide; (ii) nicotinuric acid, the glycine conjugate of nicotini...
